This invention relates to the ribonuclceotide reductase of Plasmodium falciparum (Pf RR), to the subunits of Pf RR (Pf R1 and Pf R2), and to compounds comprising peptides derived from the Pf R2 C-terminus sequence that inhibit the action of protozoal RR. The invention provides a method for the prevention and treatment of malaria caused by P,faciparum by controlling the proliferation of P. faciparum comrpising administering to a patient at least one peptide according to the invention. Antimalarial compositions are provided which comprise a pharmaceutically acceptable carrier and at least one peptide according to the invention which inhibits the P. faciparum ribonucleotide reductase reductase reduction of ribonucleotides to 2'-deoxyribonucleotides. Also provided are methods for diagnosing malaria and for screening potential antimalarial agents.
Cette invention concerne la réductase ribonucléotidique de Plasmodium falciparum (Pf RR), les sous-unités de Pf RR (Pf R1 et Pf R2) et des composés comprenant des peptides dérivés de la séquence à terminaison C de Pf R2 qui inhibent l'action du RR protozoaire. Cette invention concerne également un procédé de prophylaxie et de traitement de la malaria provoquée par P. falciparum selon lequel on régule la prolifération de P. falciparum en administrant à un patient au moins un peptide de cette invention. Des compositions antipaludiques sont décrites, ces dernières comprenant un support pharmaceutiquement acceptable et au moins un peptide de cette invention qui inhibe la réduction de la réductase ribonucléotidique de P. falciparum des ribonucléotides en 2'-désoxyribonucléotides. Cette invention concerne également des procédés de diagnostic de la malaria et de recherche d'agents antipaludiques potentiels.
