The present invention relates to polymer-modified viruses, processes for obtaining them and their use. In a preferred embodiment the polymer is polyethylene glycol (PEG). In one embodiment, the polymer is directly covalently attached to the virus. In another embodiment, the polymer is indirectly covalently attached to the virus via an intermediate coupling moiety. In yet another embodiment, the polymer is indirectly noncovalently attached to the virus via a ligand. In a preferred embodiment, the ligand has specificity for a viral surface component. For example, the ligand may be an antibody. The present invention further provides a method of making viruses modified by polymers, whereby the modified viruses retain infectivity. Another embodiment of the present invention provides a method for introducing a transgene into a target cell comprising contacting the target cell with a polymer-modified virus, wherein the virus comprises the transgene. The present invention further provides a method of delivering a virus to a tumor, comprising administering a polymer-modified virus of the invention to a subject in need of such treatment under conditions whereby the polymer- modified virus localizes to a tumor. In another embodiment, the present invention provides a composition comprising a virus modified by a polymer and a carrier.
La présente invention concerne concerne des virus modifiés par des polymères, leur procédé d'obtention ainsi que leur utilisation. Dans un mode de réalisation préféré le polymère est du polyéthylène glycol (PEG). Dans un mode de réalisation, le polymère est fixé directement de manière covalente au virus. Dans un autre mode de réalisation, le polymère est fixé indirectement de manière covalente au virus par une fraction de couplage intermédiaire. Dans un autre mode de réalisation, le polymère est fixé indirectement de manière non covalente au virus par un ligand. Dans un mode de réalisation préféré, le ligand présente une spécificité vis-à-vis d'un constituant de surface virale. Par exemple, le ligand peut être un anticorps. La présente invention concerne également un procédé de production de virus modifiés par des polymères, selon lequel les virus modifiés conservent leur pouvoir infectant. Un autre mode de réalisation de l'invention concerne une méthode d'introduction d'un transgène dans une cellule cible consistant à mettre la cellule cible en contact avec un virus modifié par polymère, le virus comprenant le transgène. En outre, la présente invention concerne une méthode d'administration d'un virus à une tumeur, consistant à administrer un virus modifié par polymère de l'invention à un sujet nécessitant un tel traitement, dans des conditions dans lesquelles le virus modifié par polymère est localisé sur une tumeur. Dans un autre mode de réalisation, la présente invention concerne une composition comprenant un virus modifié par un polymère et un excipient.
