Methods are provided for the stabilization of proteinases by the covalent attachment of or admixture with water-soluble polymers. The resultant stabilized proteinases have increased stability under the harsh conditions used in industrial genomics, which permits their use in the extraction and isolation of nucleic acids and the identification of disease-related prion proteins at elevated temperatures in solutions containing chaotropic agents, such as sodium dodecyl sulfate, urea or guanidinium salts, conferring advantages for robotic applications.
L'invention concerne des procédés permettant la stabilisation de protéinases par la fixation covalente de polymères solubles dans l'eau ou par ajout à ces polymères solubles dans l'eau. Les protéinases ainsi stabilisées ont une stabilité accrue dans les conditions difficiles appliquées en génomique industrielle, ce qui permet leur utilisation pour extraire et isoler les acides nucléiques et identifier les protéines prion liées à des maladies à des températures élevées dans des solutions contenant des agents chaotropiques, tels que du sulfate dodécyl de sodium, de l'urée ou des sels de guanidinium, avantageux pour des applications robotisées.
