The present invention relates to a polymyxin derivative wherein R1, R2 and R3 are optional and R1, R2, R3, R5, R8 and R9 are cationic or neutral amino acid residues selected so that the total number of positive charges at physiological pH is at least two but no more than three; and to a combination product comprising at least two such derivatives. The invention further relates to a method for treating, alleviating or ameliorating an infection in a subject, caused by a Gram-negative bacterium by administering a therapeutically effective amount of a derivative according to the present invention to said subject; to a method for sensitizing Gram-negative bacteria to an antibacterial agent by administering, simultaneously or sequentially in any order a therapeutically effective amount of said antibacterial agent and a derivative according to the present invention to said subject; to methods for developing novel antibiotics; for reducing the nephrotoxicity, for improving the pharmacokinetic properties of natural polymyxins and octapeptins; and for sensitizing clinically important bacteria to a host defence mechanism complement present in serum. Finally, the invention relates to a process for preparing such polymyxin derivatives.
La présente invention concerne un dérivé de polymyxine dans lequel R1, R2 et R3 sont facultatifs et R1, R2, R3, R5, R8 et R9 sont des résidus acides aminés cationiques ou neutres choisis de façon à ce que le nombre total de charges positives au pH physiologique soit au moins égal à deux mais non supérieur à trois ; et un produit combiné comprenant au moins deux de ces dérivés. L'invention concerne en outre une méthode permettant de traiter, soulager ou améliorerguérir chez un sujet une infection provoquée par une bactérie à Gram négatif en administrant audit sujet une quantité thérapeutiquement efficace d'un dérivé selon la présente invention ; une méthode de sensibilisation de bactéries à Gram négatif à un agent antibactérien en administrant audit sujet, simultanément ou successivement dans un ordre quelconque, une quantité thérapeutiquement efficace dudit agent antibactérien et d'un dérivé selon la présente invention ; des méthodes de développement de nouveaux antibiotiques, de réduction de la néphrotoxicité, d'amélioration des propriétés pharmacocinétiques de polymyxines et d'octapeptines naturelles et de sensibilisation de bactéries cliniquement importantes au complément d'un mécanisme de défense d'un hôte présent dans le sérum. Enfin, l'invention concerne un procédé de préparation de ces dérivés de polymyxine.
