A novel nucleic acid construct for down-regulating angiogenesis in a tissue of a subject is provided. The nucleicacid construct includes: (a) a first polynucleotide region encoding a chimeric polypeptide including a ligand binding domain fused to an effector domain of an apoptosis signaling molecule; and (b) a second polynucleotide region encoding a cis acting regulatory element being for directing expression of the chimeric polypeptide in a specific tissue or cell; wherein the ligand binding domain is selected such that it is capable of binding a ligand present in the specific tissue or cell, whereas binding of the ligand to the ligand binding domain activates the effector domain of the apoptosis signaling molecule. Also provided are methods of utilizing this nucleic acid construct for treating diseases characterized by excessive or aberrant neo-vascularization or cell growth.
La présente invention concerne une nouvelle construction d'acide nucléique destinée à la régulation négative de l'angiogenèse dans le tissu d'un sujet. La construction d'acide nucléique précitée comprend : (a) une première région de polynucléotide qui code un polypeptide chimérique comprenant un domaine de liaison de ligand fusionné à un domaine effecteur d'une molécule de signalisation d'apoptose; et (b) une seconde région de polynucléotide qui code un élément régulateur agissant en cis destiné à diriger l'expression du polypeptide chimérique dans un tissu ou une cellule spécifique; le domaine de liaison de ligand étant choisi de façon qu'il est capable de lier un ligand présent dans la cellule ou le tissu spécifique, alors que la liaison du ligand au domaine de liaison de ligand active le domaine effecteur de la molécule de signalisation d'apoptose. L'invention se rapporte également à des procédés d'utilisation de la construction d'acide nucléique précitée dans le traitement de maladies caractérisées par une néovascularisation ou une croissance cellulaire excessive ou aberrante.
