The invention provides an isolated polypeptide, or a fragment thereof, comprising a coiled-coil and an engineered site sufficient for the addition of a "moiety", i.e., a group, that is one or more of a phosphate, ubiquitin, glycosyl or ADP-ribosyl moiety, wherein the polypeptide binds to a binding partner in a phosphorylation-, ubiquitination-, glycosylation- or ADP- ribosylation-dependent manner. The invention also relates to methods and kits utilizing an isolated polypeptide and its binding partner, which methods and kits permit monitoring of addition or removal of the one or more moieties.
L'invention se rapporte à un polypeptide isolé, ou à un fragment de polypeptide, présentant une structure bispiralée et comportant un site construit permettant l'addition d'une "fraction", c'est à dire d'un groupe, qui est composée d'une ou plusieurs fractions phosphate, ubiquitine, glycosyle ou ADP-ribosyle, ledit polypeptide se liant à un partenaire de liaison d'une manière liée au déroulement d'une phosphorylation, d'une ubiquitination, d'une glycosylation ou d'une ADP-rybosylation. L'invention se rapporte également à des procédés et à des trousses mettant en oeuvre un polypeptide isolé et son partenaire de liaison et permettant la surveillance de l'addition ou du retrait d'une ou de plusieurs fractions.
