The present invention relates to polypeptides having phytase activity. These polypeptides have an amino acid sequence which has at least 70% identity to either of three phytases derived from the bacterium Buttiauxella, and which comprises at least one of the following amino acids at the position indicated: 119N, 120L, and/or 121E. These phytases have an improved specific activity. Additional specific amino acid substitutions are also disclosed which characterize and distinguish additional phytases of the invention having improved properties such as temperature and/or pH stability, pH activity profile, temperature activity profile, substrate profile, improved performance in animal feed in vitro orin vivo. The invention also relates to isolated polynucleotides encodingthe polypeptides, nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods for producing and using the polypeptides.
La présente invention porte sur des polypeptides ayant une activité phytase. Ces polypeptides ont une séquence d'acides aminés qui a au moins 70 % d'identité avec l'un ou l'autre de trois phytases issues de la bactérie Buttiauxella, qui compporte au moins l'un des acides aminés suivants dans la position indiquée : 119N, 120L et/ou 121E. Ces phytases ont une activité spécifique améliorée. Des substitutions supplémentaires d'acides aminés spécifiques sont également décrites, qui caractérisent et distinguent les phytases supplémentaires de l'invention ayant des propriétés améliorées telles que la stabilité à la température et/ou au pH, le profil d'activité selon le pH, le profil d'activité selon la température, le profil de substrat, l'efficacité améliorée dans la nourriture animale in vitro ou in vivo. L'invention porte également sur des polynucléotides isolés codant pour les polypeptides, les produits de construction d'acide nucléique, les vecteurs et les cellules hôtes comportant les polynucléotides, ainsi que sur des procédés de fabrication et d'utilisation des polypeptides.
