Abstract of the Disclosure Methods for synthesis and manufacture of polysaccharide-protein conjugate vaccines at high yield are provided. The methods involve reaction of a hydrazide group on one reactant with an aldehyde or cyanate ester group on the other reactant. The reaction proceeds rapidly with a high conjugation efficiency, such that a simplified purification process can be employed to separate the conjugate product from the unconjugated protein and polysaccharide and other small molecule by-products.

L'invention concerne des méthodes de synthèse et de production de vaccins conjugués polysaccharide-protéine à haut rendement. Les méthodes consistent à mettre en réaction un groupe hydrazide placé sur un réactif avec un groupe aldéhyde ou ester de cyanate placé sur l'autre réactif. La réaction s'effectue rapidement avec une très grande efficacité de conjugaison, si bien qu'un processus de purification simplifié peut être mis en oeuvre pour séparer le produit conjugué de la protéine et du polysaccharide non conjugués et des autres sous-produits à petites molécules.
