High capacity drug reservoirs are provided for incorporation into transdermal drug delivery systems. The drug reservoirs are hydrogels formulated from polyurethanes crosslinked with diisocyanate crosslinking agents or cured with radiation in the presence of a photoinitiator. Drug loading as high as 65 to 70 wt.% or higher can be achieved by absorbing drug formulation into the reservoir after hydrogel synthesis. Methods for making and using transdermal systems containing such reservoirs are provided as well.
L'invention porte sur des réservoirs à médicament de grande capacité destinés à être incorporés à des dispositifs d'administration transdermique de médicament. Ces réservoirs sont des hydrogels formulés à partir de polyuréthanes réticulés par des agents de réticulation de diisocyanate ou durcis sous l'effet d'un rayonnement en présence d'un photoamorceur. L'absorption de la formulation pharmaceutique par le réservoir après la synthèse de l'hydrogel permet le chargement d'un médicament dans un volume égal ou supérieur à 65 à 70 % en poids. L'invention porte également sur des procédés de fabrication de dispositifs transdermiques comportant ces réservoirs ainsi que sur leur utilisation.
