The present invention relates to an isolated insulator DNA molecule consisting of the 5' and 3' regions of the porcine wap gene locus and a method for insulating the expression of an introduced heterologous gene from silencing and variegation effect in chromatin into which the gene has integrated. The invention is also directed to mammalian cells comprising said insulator DNA molecule and non human animals obtained therefrom, which are preferably able to express a polypeptide of interest in milk.
L'invention concerne une molécule d'ADN d'isolateur isolée, constituée des régions 5' et 3' du locus de gène wap porcin, et un procédé permettant d'isoler l'expression d'un gène hétérologue introduit contre un effet d'extinction de l'expression et de variégation se produisant dans la chromatine dans laquelle le gène est intégré. L'invention a également trait à des cellules de mammifère contenant cette molécule d'ADN d'isolateur, ainsi qu'à des animaux non humains obtenus à partir de celles-ci, qui sont de préférence capables d'exprimer un polypeptide voulu dans le lait.
