The present invention concerns a poxviral particle having a targeted infection specificity conferred by an heterologous ligand moiety present at the surface of said poxviral particle and capable of specifically recognizing and binding to an anti-ligand molecule localized at the surface of target cells. The present invention further relates to a vector comprising a nucleotide sequence encoding a chimeric polypeptide including such an heterologous ligand moiety and all or part of a natural poxviral surface polypeptide. The present invention additionally concerns compositions comprising said poxviral particle or said vector as well as their use for therapeutic and prophylactic purposes. The invention is of very special interest in gene therapy applications, in particular in preventing or treating cancer in mammals.
La présente invention porte sur une particule poxvirale induisant une infection en particulier causée par un ligand hétérologue présent à la surface de ladite particule poxvirale et capable de reconnaître précisément une molécule anti-ligand située à la surface de cellules cibles et de s'y lier. La présente invention se rapporte également à un vecteur constitué d'une séquence nucléotidique codant un polypeptide chimère comprenant ce ligand hétérologue et un polypeptide naturel situé à la surface de la particule poxvirale en totalité ou en partie. La présente invention concerne aussi des compositions contenant ladite particule poxvirale ou ledit vecteur, de même que leur utilisation à des fins thérapeutiques et prophylactiques. L'invention est particulièrement pertinente dans des applications de la thérapie génique, surtout pour prévenir ou traiter le cancer chez les mammifères.
