The invention relates to a method for the preparation of efficient antigen- presenting human .gamma..delta. T cells, to the .gamma..delta. T cells prepared by such a method, and to their use in immunotherapy, vaccination, vaccine development and diagnostics. Similar to dendritic cells (DCs) in potency and efficacy, these human .gamma..delta. T cells process antigens and present antigenic peptides to .alpha..beta. T cells and induce antigen- specific responses (proliferation and differentiation) in naïve .alpha..beta. T cells. .gamma..delta. T cells are easily purified from peripheral blood, acquire "maturation" status (expression of essential adhesion, co-stimulatory and major histocompatibility complex molecules) within 1 day of in vitro culture under stimulation and induce strong primary and secondary T helper cell and cytotoxic T cell responses. The .gamma..delta. T cells may be used in a method of treatment of tumors or chronic or recurrent infectious diseases, in identification of novel tumor or pathogen-derived antigens, and in the diagnosis of the immune competence of a patient.
La présente invention a trait à un procédé pour la préparation de cellules T .gamma..delta. humaines efficaces présentatrices de l'antigène, à des cellules T .gamma..delta. préparées par un tel procédé, et à leur utilisation en immunothérapie, vaccination, développement de vaccins et diagnostic. Semblables aux cellules dendritiques en puissance et efficacité, ces cellules T .gamma..delta. humaines assurent le traitement des antigènes et présentent des peptides antigéniques aux cellules T .alpha..beta. et l'induction de réponses spécifiques de l'antigène (prolifération et différenciation) dans des cellules T .alpha..beta. naïves. Les cellules T .gamma..delta. sont facilement purifiées à partir du sang périphérique, acquièrent un statut de maturation (expression d'adhésion essentielle, molécules complexes co-stimulatrices et d'histocompatibilité majeure) en l'espace d'un jour de culture in vitro sous stimulation et induisent de fortes réponses de lymphocytes T auxiliaires et de lymphocytes T cytotoxiques primaires et secondaires. Les cellules T .gamma..delta. peuvent être utilisées dans un procédé de traitement de tumeurs ou de maladies infectieuses chroniques et récurrentes, dans l'identification de nouveaux antigènes tumoraux ou dérivés d'agents pathogènes, et dans le diagnostic de la compétence immune d'un patient.
