The invention relates to method for the preparation of metal oxide supports loaded with biomolecules, comprising the steps of: (a) activating the surface of the support by means of a silanating agent comprising an amine group; (b) loading the support by attaching biomolecules to the activated surface, characterized in that subsequently the loaded support is treated with an acidic solution, and provided that the method is not used for the preparation of silica wafers which are aminated by silanation using (3- aminoprdeyl)monoethoxydimethylsilane and loaded with oligonucleotides. Similarly, an activated and loaded support may be treated with a basic or neutral solution, provided that the method is not used for derivatization of aluminiumoxide nanoparticles aminated with (3-aminopropyl)triethoxysilane, wherein the basic solution further contains a large excess of N- acetylhomocysteinelactone. This method can for example be used in the preparation of arrays for probe-based assays.
L'invention concerne un procédé de préparation de supports d'oxydes métalliques chargés de biomolécules, qui consiste à: (a) activer la surface du support par un agent de silanation comprenant un groupe amine; (b) charger le support en fixant des biomolécules à la surface activée, le support étant par la suite traité par une solution acide, pour autant que le procédé ne serve pas à la préparation de plaquettes en silicium aminées par silanation au moyen de (3-aminopropyl)monoéthoxydiméthylsilane et chargées d'oligonucléotides. De même, un support activé et chargé peut être traité par une solution basique ou neutre, pour autant que le procédé ne serve pas à la dérivation de nanoparticules d'oxyde d'aluminium aminées par (3-aminopropyl)triéthoxysilane, la solution basique contenant en outre N-acétylhomocystéinelactone en large excès. On peut utiliser le procédé pour, par exemple, préparer des réseaux de dosages par sondes.
