The present invention is directed to processes for the synthesis of morphinans. In particular, a process for the asymmetric reduction of an imine moiety in a 3,4-dihydroisoquinoline to produce a tetrahydroisoquinoline, followed by a Birch reduction to produce a hexahydroisoquinoline. In various embodiments, the 3,4-dihydroisoquinoline contains a phenol moiety protected with a labile protecting group. In other embodiments, the imine reduction reaction mixture contains silver tetrafluoroborate.

La présente invention concerne des procédés de synthèse de morphinanes. En particulier, la présente invention concerne un procédé de réduction asymétrique d'un fragment imine dans une 3,4-dihydroisoquinoléine de façon à produire une tétrahydroisoquinoléine, suivie d'une réduction de Birch de façon à produire une hexahydroisoquinoléine. Dans divers modes de réalisation, la 3,4-dihydroisoquinoléine contient un fragment phénol protégé par un groupe protecteur labile. Dans d'autres modes de réalisation, le mélange réactionnel de réduction d'un imine contient le tétrafluoroborate d'argent.
