The present invention relates to substances capable of inhibiting presenilin 2 expression in neurodegenerative diseases and in Alzheimer's disease. Ribozymes capable of cleaving presenilin 2-specific RNA are disclosed. Preferably, the invention concerns fusion ribozymes comprising a presenilin 2-specific ribozyme and an autocatalytical hammerhead ribozyme. Furthermore, recombinant DNA molecules coding for said ribozymes, a recombinant vector comprising the cDNA corresponding to said ribozymes and a host cell comprising said recombinant vector are disclosed. Additionally, the invention pertains to pharmaceutical compositions comprising said substance or ribozyme or recombinant vector and a pharmaceutically acceptable carrier. The invention also comprises the use of said substance or ribozyme or recombinant vector for the treatment of neurodegenerative diseases or for the treatment of Alzheimer's disease.
La présente invention concerne des substances permettant d'inhiber l'expression de la préséniline 2 dans les maladies neurodégénératives et dans la maladie d'Alzheimer. L'invention traite aussi des ribozymes pouvant scinder l'ARN spécifique de la préséniline 2. De préférence, l'invention a pour objet des ribozymes de fusion comprenant un ribozyme spécifique de la préséniline 2 et un ribozyme marteau autocatalytique. En outre, des molécules d'ADN de recombinaison codant lesdits ribozymes, un vecteur de recombinaison comprenant l'ADNc correspondant auxdits ribozymes et à une cellule hôte comprenant ledit vecteur de recombinaison sont décrits. En outre, l'invention traite des compositions pharmaceutiques comprenant ladite substance ou le ribozyme ou le vecteur de recombinaison et un support pharmaceutiquement acceptable. L'invention traite également de l'utilisation de ladite substance ou du ribozyme ou du vecteur de recombinaison pour traiter les maladies neurodégénératives ou pour traiter la maladie d'Alzheimer.
