The present invention provides a method of assaying for and arresting, preventing and/or reversing the impairment of central and peripheral nervous system function comprising reducing .beta.-amyloid plaque burden by the administration of compounds that reduce apoE expression. The compounds used in the method of the invention may be: 1) inhibitors of 3-hydroxy-3- methylglutaryl coenzyme A (HMG CoA) reductase; 2) inhibitors of cholesterol biosynthesis; 3) inhibitors of protein isoprenylation, specifically geranylgeranylation; and/or 4) inhibitors of NF-~B activation or function. Assays for compounds with inhibit apoE expression from microglial cells are also disclosed.

L'invention concerne une méthode d'analyse permettant de stopper, de prévenir et/ou d'inverser la dégénérescence des fonctions des systèmes nerveux central et périphérique. Cette méthode consiste à réduire la plaque .beta.-amyloïde par l'administration de composés qui réduisent l'expression d'apoE. Les composés utilisés dans la méthode selon l'invention peuvent être: 1) des inhibiteurs de 3-hydroxy-3-méthylglutaryle coenzyme A (HMG CoA) réductase; 2) des inhibiteurs de la biosynthèse du cholestérol; 3) des inhibiteurs d'isoprénylation de protéines, notamment de géranylgéranylation de protéines; et/ou 4) des inhibiteurs de l'activation ou de la fonction de NF-~B. L'invention concerne également des analyses permettant d'obtenir des composés inhibant l'expression d'apoE dans les microglies.
