The invention relates to the generation of Fusion-Related Molecular Structures (FRMS) which comprise one or more transitional fusion-related determinants. Highly effective vaccines may be constructed which present the fusion-related determinants to the immune system that engender unique antibodies capable of potently neutralizing a broad range of primary isolates, from worldwide locations and from different phylogenetic viral clades. The present invention provides methods for formation, isolation and purification of the FRMS as well as the use of such FRMS in a variety of compositions and methods, including, for example, as vaccine immunogens, diagnostics, and therapeutics. The present invention concerns FRMS capable of eliciting neutralizing antibodies to viral pathogens and primary isolates of a virus. Antibodies raised to the FRMS can be used to study the molecular pathway toward fusion of the virus and host cell and to identify points of a possible antibody-mediated blockade to that pathway. The invention also relates to the use of antibodies of the invention for anti-viral agents, blood product additives, contraceptive additives passive immunization in post-exposure treatments or fetus immunization.

La présente invention concerne la génération de structures FRMS (Fusion-Related Molecular Structures) comprenant un ou plusieurs déterminants transitionnels concernés par la fusion. On peut construire des vaccins très efficaces et qui présentent les déterminants concernés par les fusions au système immunitaire qui engendre des anticorps uniques capables de neutraliser puissamment une large gamme d'isolats primaires, et ce, avec malgré des provenances mondiales même à partir de différentes clades virales phylogénétiques. L'invention concerne des procédés permettant de former, d'isoler et de purifier la FRMS ainsi que l'utilisation de telles FRMS dans une variété de compositions et procédés, et notamment comme immunogènes vaccinaux, comme agents diagnostics et comme agents thérapeutiques. L'invention concerne notamment des FRMS capables de mettre en évidence la neutralisation d'anticorps face à des pathogènes viraux et des isolats primaires d'un virus. Des anticorps dirigés contre les FRMS peuvent servir à l'étude du chemin moléculaire aboutissant à la fusion du virus avec la cellule hôte, et à l'identification de points d'un possible blocage à médiation d'anticorps survenant sur ce chemin. L'invention concerne enfin l'utilisation d'anticorps de l'invention comme agents antiviraux, comme additifs pour produits sanguins, comme agent d'immunisation passive pour additifs contraceptifs dans le traitement postexposition ou l'immunisation foetale.
