From a human antibody phage library, a clone BM095, which shows a potent proliferation inhibitory activity in an IL-31-dependent Ba/F3 cell proliferation assay system, is obtained. When administered to an atopic dermatitis model animal with the use of an Nc/Nga mouse, a model mouse of chronic dermatitis induced by the repeated application of picryl chloride, a rheumatoid arthritis model mouse and an arthritis deformans model mouse, this antimouse NR10 neutralizing antibody shows a remarkable effect of inhibiting the symptoms. Thus, it is clarified that this anti-NR10 neutralizing antibody is useful in practice as a remedy for inflammatory diseases. Moreover, antihuman NR10 neutralizing antibody is successfully acquired and thus a highly useful remedy which is clinically applicable in practice is provided.
La présente invention concerne un clone, BM095a, tiré d'une banque de phages anticorps humains, qui présente une puissante activité inhibitrice de la prolifération dans un système de dosage de la prolifération de cellules Ba/F3 dépendante d'IL-31. Selon l'invention, cet anticorps neutralisant anti-NR10 de souris présente un effet remarquable d'inhibition des symptômes quand il est administré à un modèle animal de la dermatite atopique en utilisant une souris Nc/Nga, un modèle souris de dermatite chronique induite par l'application répétée de chlorure de picryle, un modèle souris de l'inflammation articulaire rhumatoïde et un modèle souris de la polyarthrite rhumatoïde. Il est donc confirmé cet anticorps neutralisant anti-NR10 est utilisable en pratique pour soigner les maladies inflammatoires. D'autre part, il est facile d'obtenir un anticorps neutralisant anti-NR10 humain et l'invention concerne donc un remède très utile cliniquement applicable en pratique.
