PROCEDURE FOR DETECTING BETA-HEMOLYTIC STREPTOCOCCUS ANTIGENS Abstract of the Disclosure A method for detecting beta-hemolytic streptococcus antigens in clinical specimens employing nitrous acid extrac- tion and latex agglutination techniques is disclosed. In the practice of the invention, the antigen is extracted from an uncentrifuged clinical specimen by means of nitrous acid, the extracted specimen is adjusted to an approximately neutral pH and is brought into contact with an appropriate latex aggluti- nation reagent. In preferred practice the specimen is col- lected on a throat swab and the entire extraction and neutral- ization steps are effected in a microtube in the presence of the swab. The swab is then employed to remove the neutral- ized, extracted specimen to a slide or the like onto which a few drops of the specimen are expressed by rolling the swab. Detection of the antigen by conventional latex agglutination techniques may then be accomplished optimally using latex spheres onto which high titer rabbit gamma globulin is pas- sively adsorbed.
