Process for detecting and quantifying diarrhoetic shellfish poison (DSP) based on the inhibition of phosphatases. The presence of DSP-producing red waters contaminates the molluscs which have to be constantly controlled before they are commercialised. The present technique is the bioassay which comprises the administration of a mollusc extract to rats; the result is positive if the rat dies before twelve hours. Said method is slow, costly, erratic and lacks specificity. The process of the invention detects and quantifies the toxines by measuring the inhibition of phosphatases partially purified. The phosphate inhibition is quantified by using as support a microtitration plate, and eliciting the enzymatic activity with a phosphatase substrate which, according to its chemical characteristics, is converted into a fluorescent, luminescent or colour product. Said technique is fast, economical and reliable. The sector which can beneficiate from this invention is the mollusc canning and producing industry.
L'invention concerne le procédé permettant de détecter et de quantifier des toxines diarrhéiques des dinoflagellés en ayant recours à l'inhibition de phosphates. La présence de marées rouges productrices desdites toxines contamine les mollusques qui doivent être contrôlés en permanence avant d'être commercialisés. La technique actuelle est le bioessai qui consiste à administrer un extrait de mollusque à des rats; le résultat est positif si le rat meurt avant qu'un laps de temps de douze heures se soit écoulé. Ce procédé est lent, coûteux, erratique et peu spécifique. Le procédé de l'invention permet de détecter et de quantifier les toxines en mesurant l'inhibition des phosphatases partiellement purifiées. L'inhibition de phosphatases est quantifiée en utilisant comme support une plaque de microtitrage et en revelant l'activité enzymatique à l'aide d'un substrat de phosphatase qui, selon ses caractéristiques chimiques, se convertit en un produit fluorescent, luminescent ou coloré. Cette technique est rapide, économique et fiable. Le secteur pouvant bénéficier de ce procédé est l'industrie de production et de mise en conserve de mollusques.
