A method of concentrating a disease-related conformation of a protein such as the PrPSc in a sample is disclosed. The method comprises liquefying the sample and adding a complexing agent such as phosphotungstic acid (PTA) which complexes preferentially or exclusively with the PrPSc. After the complex is formed the composition is centrifuged until the complex settles at the bottom. Thereafter, the supernatant is poured away. The remaining pellet may be suspended in an aqueous solution containing a protease inhibitor for storage. The PTA stains the PrPSc making the resulting concentrated PrPSc susceptible to further analysis, making it possible to quickly and efficiently determine the presence of PrPSc and its concentration in a sample. The method can be used to render a sample non-infectious by removing all or substantial of the infectious form of a protein from a sample.
Cette invention a trait à une technique permettant de concentrer une protéine à conformation liée à une affection, notamment le prion Sc (PrP?Sc¿) présent dans un prélèvement. Cette technique consiste à liquéfier le prélèvement et à y ajouter un agent de complexion, un acide phosphotungstique (PTA), formant, de manière préférentielle ou exclusive, un complexe avec le PrP?Sc¿. Une fois le complexe formé, on soumet la composition à une centrifugation jusqu'à ce que le complexe occupe le fond du contenant. Après cela, on évacue le liquide surnageant. Le sédiment restant peut être remis en suspension dans une solution aqueuse contenant un inhibiteur de protéase aux fins du stockage. Le PTA colore le PrP?Sc¿ faisant mieux se prêter le PrP?Sc¿ concentré résultant à une analyse ultérieure et rendant possible une détermination rapide et efficace de la présence du PrP?Sc¿ et de sa concentration dans le prélèvement. Il est également possible d'utiliser cette technique pour rendre un prélèvement non infectieux et ce, en éliminant toute forme infectieuse ou une grande partie de celle-ci d'une protéine dans un prélèvement.
