The invention pertains to a process for extracting and buffering and/or concentrating dissolved macromolecules of a mac- romolecular mixture using a liquid chromatography apparatus, wherein buffer solutions that promote the separation process are added to the macromolecular mixture to be separated prior to its entering into the liquid chromatography apparatus or during the separation process in the liquid chromatography apparatus, said buffer solutions then being entirely or partially withdrawn and/ or exchanged after the separation for purposes of concentrating or purifying the kind of molecules extracted. To be able to carry out such a process of buffering and/or concentrating more quickly and continuously with less outlay for equipment, the invention proposes that the buffer solution be withdrawn and/or exchanged by means of countercurent dialysis, which is carried out con- tinuously with the eluate of the liquid chromatography apparatus. The countercurrent dialysis proposed as per the invention makes it possible to supply the eluate with specific low-molecular substrates for the enzymatic detection.
