The subject invention involves processes for making 2,4-difluoro-3-Q1-benzoic acid, wherein Q1 is derived from an electrophilic reagent, comprising the steps of: (a) treating 1-bromo-2,4-difluorobenzene with a strong, non- nucleophilic base; then treating with an electrophilic reagent which provides Q1, or a functional moiety which is then transformed to Q1, producing 1-bromo- 2,4-difluoro-3-Q1-benzene; (b) treating the 1-bromo-2,4-difluoro-3-Q1-benzene with an alkali or alkaline earth metal or organometallic reagent; then treating with carbon dioxide, or with a formylating agent followed by oxidation, to produce 2,4-difluoro-3-Q1-benzoic acid. The subject invention also involves optional additional steps to further modify Q1, or to substitute a non-hydrogen moiety at the 5-position, at the 6-position, or at both, of the phenyl ring of the 2,4-difluoro-3-Q1-benzoic acid.
La présente invention concerne des procédés permettant de préparer de l'acide 2,4-difluoro-3-Q1-benzoïque, Q1 étant dérivé d'un réactif électrophile. Les procédés de cette invention consistent: a) à traiter du 1-bromo-2,4-difluorobenzène avec une base forte non nucléophile, puis avec un réactif électrophile apportant Q1 ou un fragment fonctionnel transformé ensuite en Q1, de manière à produire du 1-bromo-2,4-difluoro-3-Q1-benzène; b) à traiter le 1-bromo-2,4-difluoro-3-Q1-benzène ainsi obtenu avec un alcali, un métal alcalino-terreux, ou un réactif organométallique, puis avec du dioxyde de carbone ou un agent de formylation, ce traitement étant suivi d'une oxydation de manière à préparer de l'acide 2,4-difluoro-3-Q1-benzoïque. La présente invention concerne également d'autres étapes permettant éventuellement de modifier Q1, ou de substituer un fragment non hydrogène en position 5, en position 6, ou les deux, du cycle phényle de l'acide 2,4-difluoro-3-Q1-benzoïque.
