A process for preparing pliable soft tissue specimen which are resistant to cracking and devoid of viable cells includes the steps of treating native soft tissue obtained from a donor by a gradually increasing gradient of aliphatic alcohol or other suitable water miscible polar organic solvent until the last alcohol (or other solvent) solution has at least 25 % by volume of the organic liquid. Thereafter, the tissue specimen is treated with a solution containing glycerol or low molecular weight ( < 1000D) polyethylene glycol, and polyethylene glycol of a molecular weight between approximately 6,000 to 15,000 D and heparin. Thereafter, the tissue specimen is briefly immersed in aqueous heparin solution, frozen and lyophilized. The tissue specimen is suitable for implantation as a homograft or xenograft, with or without rehydration.

L'invention porte sur un procédé de préparation d'échantillons de tissus souples, pliables, résistant aux craquelures et dépourvus de cellules viables. Ce procédé consiste à traiter un tissu souple natif prélevé chez un donneur par un gradient augmentant progressivement d'un alcool aliphatique ou autre solvant organique polaire approprié, miscible dans l'eau, jusqu'à ce que la dernière solution alcoolique (ou autre solvant) possède au moins 25 % en volume de liquide organique. Puis, l'échantillon de tissu est traité avec une solution contenant un glycérol ou un polyéthylèneglycol de faible poids moléculaire (<1000D), et un polyéthylèneglycol dont le poids moléculaire est compris approximativement entre 6000 et 15000 D et de l'héparine. L'échantillon de tissu est alors plongé rapidement dans une solution d'héparine aqueuse, congelée et lyophilisée. L'échantillon de tissu est approprié pour être implanté sous forme d'allogreffe ou de xénogreffe, avec ou sans réhydratation.
