Processes for the efficient preparation of 2-[R'C(O)OCH2]-1,3-dioxolanyl-5-one compounds are provided that comprise reacting an acetal of the formula (R1O)2CHR wherein R is -(CH2-O-C(O)R1), and each R1 is independently alkyl, aryl, heteroaryl, heterocyclic, alkaryl, alkylheteroaryl, or alkylheterocyclic, or aralkyl, with glycolic acid in an organic solvent, in the presence of a Lewis acid. The dioxolanylone compounds can be used in the preparation of 1,3-dioxolane nucleoside compounds.

On présente des procédés permettant de préparer de manière efficace des 2-¬R'C(O)OCH2|-1,3-dioxolanyl-5-ones, procédés consistant à faire réagir un acétal de formule (R1O)2CHR, où R est un -(CH2-O-C(O)R1), et où les R1 sont, indépendamment les uns des autres, un alkyle, un aryle, un hétéroaryle, un hétérocycle, un alkaryle, un alkylhétéroaryle, ou un alkylhétérocycle, ou un aralkyle, avec de l'acide glycolique dans un solvant organique, en présence d'un acide de Lewis. Les dioxolanylones peuvent être employées dans la préparation de nucléosides 1,3-dioxolanes.
