The present invention provides a process for preparing the antiprogestational agent, 17.alpha.-acetoxy-11.beta.-[4-N,N-( dimethy-lamino)phenyl]-21-methoxy-19-norpregna-4,9-diene-3,20-dione, intermediates useful in the process, and processes for preparing such intermediates. The process of preparing the antiprogestational agent involves replacing the cyanohydrin group of the compound of formula (1) with a chloroacetyl group and a hydroxyl group; replacing the chloro group of the resulting compound with an acetoxy group; deacetylating the resulting compound; selectively ketalizing the resulting compound; selectively methylating the 21-hydroxy group of the resulting compound; reducing the 20-keto group of the resulting compound; epoxidizing the resulting compound; introducing a N,N-dimethylaminophenyl group at the 11-position and opening the epoxide; deketalizing the resulting compound; selectively oxidizing the 20-hydroxyl group to a keto group; and acetylating the resulting compound.
La présente invention se rapporte à un procédé de préparation d'un agent antiprogestatif, la 17 alpha -acétoxy-11 beta -[4-N,N-(diméthylamino)phényl]-21-méthoxy-19-norpregna-4,9-diène-3,20-dione, à des intermédiaires utilisés par ce procédé et à des procédés de préparation de ces intermédiaires. Ledit procédé de préparation de l'agent antiprogestatif consiste à remplacer le groupe cynohydrine du composé représenté par la formule (1) par un groupe chloroacétyle et un groupe hydroxyle; à remplacer le groupe chloro du composé résultant par un groupe acétoxy; à désacétyler le composé résultant; à former sélectivement un cétal à partir du composé résultant; à méthyler sélectivement le groupe 21-hydroxy du composé résultant; à réduire le groupe 20-céto du composé résultant; à époxyder le composé résultant; à introduire un groupe N,N-diméthylaminophényle en position-11 et à ouvrir l'époxyde; à dé-cétaliser le composé résultant; à oxyder sélectivement le groupe 20-hydroxyle en groupe céto et à acétyler le composé résultant.
