Abstract The invention relates to a new process for preparing recombinant IFN-.alpha.. Expression in E. coli is carried out under the control of a phoA promotor. By linking the IFN-.alpha. gene with the STII signal sequence, the secretion of the protein into the periplasmic space and a correctly processed N-terminus are obtained. Purification of the protein is carried out by adsorption chromatography on silica gel, hydrophobic interaction chromatography, cation and anion exchange chromatography.
