A method of phosphorodiamidite production comprising the steps of reacting a phosphorus trihalide with a dialkyl amine in a polar solvent to form an intermediate compound. This intermediate compound is then subsequently reacted with an hydroxyalkyl compound and a dialkyl amine in the presence of a non- polar co-solvent. Following filtration to remove the solid by-product the two solvents form separate layers. This is advantageous as the upper, non-polar solvent, layer contains the high-purity phosphorodiamidite product.
L'invention concerne un procédé de production de phosphorodiamidite qui comprend les étapes consistant à faire réagir un trihalogénure phosphoreux avec une dialkylamine dans un solvant non polaire afin que soit un formé un composé intermédiaire. On fait ensuite réagir ce composé intermédiaire avec un composé hydroxyalkyle et une dialkylamine en présence d'un co-solvant non polaire. Après une filtration destinée à éliminer les sous-produits solides, les deux solvants forment des couches séparées. Ceci s'avère avantageux car la couche supérieure constituée par le solvant non polaire contient le produit de phosphorodiamidite de haute pureté.
