Methods for the manufacture of polysaccharide-protein conjugate vaccines at high yield are provided. The methods involve reaction of a hydrazide group on one reactant with an aldehyde group on the other reactant. The reaction proceeds rapidly with a high conjugation efficiency. Simplified purification processes can be employed to separate the conjugate product from the unconjugated protein and polysaccharide and other small molecule by-products.

L'invention concerne des procédés de préparation de vaccins conjugués polyssacharide-protéine à haut rendement. Ces procédés consistent à faire réagir un groupe hydrazide sur un réactif avec un groupe aldéhyde sur l'autre réactif. Cette réaction est rapide et présente une efficacité de conjugaison élevée. Des procédés de purification simplifiés peuvent être mis en oeuvre pour séparer le produit conjugué de la protéine et du polyssacharide non conjugués et d'autres sous-produits à petites molécules.
