A quick dissolving, high loading ribavirin composition which is essentially free of ribavirin polymorphic forms, said composition is prepared by a process comprising: (a) mixing ribavirin, at least one disintegrant and/or at least one filler to form an homogenous mixture; (b) granulating the mixture in the presence of water to form a granulation; (c) drying the granulation; and (d) mixing at least one disintegrant and/or at least one lubricant with the granulation to produce a ribavirin composition, wherein said ribavirin composition comprises at least 80 weight percent of ribavirin, based on the total weight of the composition. The ribavirin compositions prepared by the wet granulation process of the invention are free flowing and have adequate bulk density for processing into capsules. In addition, the ribavirin compositions contain at least 80 weight percent of ribavirin. Furthermore, the ribavirin compositions are substantially free of polymorphic forms of ribavirin, i.e., there are no signs of polymorphic change in ribavirin as determined by infrared spectrophotometry.
L'invention concerne une composition de ribavirine à charge élevée et à dissolution rapide, sensiblement dépourvue de formes polymorphes de ribavirine, dont le procédé de préparation consiste à : (a) mélanger de la ribavirine, au moins un délitant et/ou au moins un excipient, de façon à former un mélange homogène ; (b) granuler le mélange en présence d'eau de façon à former un produit de granulation ; (c) sécher ledit produit de granulation ; et (d) mélanger au moins un déliant et/ou au moins un lubrifiant avec ledit produit de granulation, afin de produire une composition de ribavirine, laquelle comprend au moins 80 pour cent en poids de ribavirine, par rapport au poids total de la composition. Les compositions de ribavirine préparées par le procédé de granulation humide de l'invention s'écoulent librement et présentent une masse volumique apparente appropriée pour la transformation en capsules. De plus, les compositions de ribavirine contiennent au moins 80 pour cent en poids de ribavirine. En outre, les compositions de ribavirine sont sensiblement dépourvues de formes polymorphes de ribavirine, c.-à-d., qu'il n'y a pas de signes de changement polymorphe dans la ribavirine, tels que déterminé par la spectrophotométrie infrarouge.
