A method for preparing taxane derivatives of general formula (I) by esterifying protected baccatine III or protected 10-deacetyl baccatin III with an acid of general formula (II). In general formulae (I) and (II), Ar is an aryl radical; R1 and R2 which are the same or different, are a hydrogen atom, an alkyl, phenylalkyl, phenyl, alkoxyphenyl or dialkoxyphenyl radical, or R1 and R2, together with the carbon atom to which they are attached, form a 4-7 membered ring; R3 is an acetyl radical or a hydroxy function protective grouping, and R4 is a hydroxy function protective grouping; R is a phenyl radical or a radical of formula R5-O- wherein R5- is an alkyl, alkenyl, alkynyl, cycloalkyl, cycloalkenyl, phenyl or saturated or unsaturated nitrogenous heterocylcyl radical. The products of general formula (I) are particularly useful for preparing taxol, Taxotere and analogues thereof having remarkable antitumor properties.

Procédé de préparation de dérivés du taxane de formule générale (I): (voir formule I) par estérification de la baccatine III protégée ou de la 10-désacétyl-baccatine III protégée au moyen d'un acide de formule générale (VI): (voir formule VI) Dans les formules générales (I) et (VI), Ar représente un radical aryle, R1 et R2, identiques ou différents, représentent un atome d'hydrogène, un radical alcoyle, phénylalcoyle, phényle, alcoxyphényle ou dialcoxyphényle, ou bien R1 et R2 forment ensemble avec l'atome de carbone auquel ils sont liés un cycle ayant de 4 à 7 chaînons, R3 représente un radical acétyle ou un groupement protecteur de la fonction hydroxy et R4 représente un groupement protecteur de la fonction hydroxy, R représente un radical phényle ou un radical de formule R5-O- dans laquelle R5- représente un radical alcoyle, alcényle, alcynyle, cycloalcoyle, cycloalcényle, phényle, hétérocyclyle azoté saturé ou non saturé . Les produits de formule générale (I) sont particulièrement utiles pour préparer le Taxol®, le Taxotère® et leurs analogues qui présentent des propriétés antitumorales remarquables.
