It is intended to provide a method of constructing a coffee plant with a lowered caffeine content by regulating the expression of genes encoding enzymes relating to caffeine biosynthesis, which are isolated from coffee, by the antisense method or the RNAi method. Namely, a method of constructing a caffeineless coffee plant by genetic recombination which involves the step of preparing an antisense sequence or an RNAi sequence of a gene encoding an enzyme relating to caffeine biosynthesis and thus constructing an expression vector for transformation, the step of transferring the expression vector thus obtained into an agrobacterium, the step of infecting a tissue piece wherein cell division of a coffee plant has been activated, or a callus or an adventive embryo derived from a coffee plant tissue piece with the above agrobacterium to thereby transform the tissue piece, callus or adventive embryo as described above, and the step of obtaining a transformed coffee plant from the thus transformed tissue piece, callus or adventive embryo.
L'invention concerne un procédé d'élaboration de caféier pour café à teneur réduite en caféine par régulation de l'expression de gènes codant les enzymes liées à la synthèse de la caféine, que l'on isole du café, selon une technique antisens ou ARNi. Précisément, il s'agit d'une recombinaison génétique, selon les étapes suivantes: préparation de séquence antisens ou ARNi d'un gène codant une enzyme liée à la biosynthèse de la caféine, et élaboration d'un vecteur d'expression pour la transformation; transfert de ce vecteur dans une agrobactérie; infection d'un morceau de tissu dans lequel la division cellulaire d'un caféier a été activée, ou bien d'un cal ou d'un embryon adventif de caféier, par ladite agrobactérie, aux fins de transformation (morceau de tissu, cal ou embryon), comme indiqué précédemment, pour donner le caféier transformé (par morceau de tissu, cal ou embryon transformé).
