It is intended to provide an expression system of a recombinant protein and a cell-free translation system with the use of a host wherein a target protein is surely incorporated into the stereo structure of chaperonin as a fused protein with a chaperonin subunit (namely, a molecular chaperonin of about 60 kDa, heat shock protein of 60 kDa or thermosome) to thereby inhibit the expression of toxicity of the target protein in the host, the formation of an enclosed matter, and digestion with a protease, thereby universally expressing any protein as a soluble protein. Namely, a process for producing a protein characterized by transcribing and translating a gene containing a gene encoding a chaperonin subunit and a gene encoding a target protein to thereby synthesize a fused protein in which the target protein is linked to the chaperonin subunit via a peptide bond.

L'invention concerne un système d'expression de protéine de recombinaison et un système de translation cellulaire utilisant un hôte, une protéine cible étant incorporée de manière sûre dans la structure stéréo d'une chaperonine sous forme de protéine fusionnée avec une sous-unité de chaperonine (essentiellement, une chaperonine moléculaire d'environ 60kDa, une protéine de choc thermique de 60kDa ou un thermosome), afin d'inhiber l'expression de la toxicité de ladite protéine cible dans l'hôte, la formation d'une subsistance incorporée, et la digestion par une protéase, ce qui permet d'exprimer universellement une protéine quelconque sous forme de protéine soluble. L'invention concerne également un procédé de production de protéine caractérisé par la transcription et la translation d'un gène contenant un gène qui code pour une sous-unité de chaperonine et d'un gène qui code pour une protéine cible afin d'effectuer la synthèse d'une protéine fusionnée, ladite protéine cible étant liée à la sous-unité de chaperonine via une liaison peptidique.
