Ce procédé est caractérisé en ce que: (1) on hybride la séquence sur laquelle se trouve la base à identifier avec un nucléotide de longueur suffisante; (2) on débute la synthèse du brin complémentaire de l'hybride obtenu en (1), -ledit nucléotide servant d'amorce- en présence: d'une polymérase sans action exonucléase 3'5' et d'au moins une base nucléotidique modifiée; (3) on détecte la base nucléotidique bloquante incorporée par tout moyen approprié. Application: diagnostic des maladies génétiques.
