The invention relates to a method for (pre)treating affinity chromatography matrices, preferably protein A matrices, comprising at least one surface active substance for reducing ligand leakage. Treating the affinity chromatography matrix according to the invention makes it possible to obtain a constant quality of the affinity chromatography matrix thereby representing an important requirement for the suitability/use in biopharmaceutical processes. The invention also relates to methods for determining the ligand leakage.
L'invention concerne un procédé permettant de (pré)traiter des matrices de chromatographie d'affinité, de préférence des matrices de protéine A, avec au moins une substance tensioactive pour réduire la fuite de ligands. Le traitement selon l'invention de la matrice de chromatographie d'affinité permet de parvenir à une qualité contante de la matrice de chromatographie d'affinité, en termes de fuite de ligands, ce qui constitue un préalable important pour sa qualification/son utilisation dans des processus pharmaceutiques. L'invention concerne en outre des méthodes permettant de déterminer la fuite de ligands.
