A process for the generation of oligonucleotide libraries representative of a given template is described. Starting from a random pool of oligonucleotides, the process selects only those which hybridize to the template nucleic acid. This selection yields a highly specific library that represents an oligo-image of the chosen template. The novel quality of this approach is the generation of amplifiable oligonucleotide probes that are of uniform length, free of repetitive sequence motifs and easily subjected to differential selection. This technique is used to produce different oligonucleotide libraries (OLs) and shows that these OLs do not cross-hybridize. Differential selection of these OLs produces oligonucleotides that can be used in the identification, characterization and isolation of nucleic acids.

L'invention concerne la production d'une banque d'oligonucléotides représentative d'une matrice donnée. Dans un groupe d'oligonucléotides aléatoire, on ne sélectionne que ceux qui s'hybrident au contact d'un acide nucléique matriciel. Grâce à cette sélection, on obtient une banque très spécifique représentant une oligo-image de la matrice choisie. La nouveauté de cette approche est la production de sondes d'oligonucléotides amplifiables, de longueur uniforme, exemptes de motifs séquentiels répétitifs et facilement soumises à une sélection différentielle. Cette technique s'utilise pour la production de banques d'oligonucléotides (OL) différentes et montre que ces OL ne subissent aucune hydridation croisée. La sélection différentielle de ces OL produit des oligonucléotides pouvant s'utiliser pour l'identification, la caractérisation et l'isolation d'acides nucléiques.
