A novel process for the phosphorylation of insulin in which peptide is contacted with phosphorous oxychloride in an aque- ous solvent is disclosed. Phosphorylated insulin with the ability to produce a higher degree of blood glucose control in the treat- ment of diabetes mellitus is obtained by this process. The phosphorylated insulin of the invention produces a significantly lower change in blood glucose per % change in insulin dose when compared to presently available insulin.
