Abstract of the disclosure: The invention provides a process for the preparation of human insulin or the derivatives thereof from pig insulin or the derivatives thereof, which comprises re- acting pig insulin or a derivative thereof at a pH below its isoelectric point with an excess of a threonine ester or one of its derivatives in the presence of trypsin or a trypsin-like enzyme, and optionally splitting off the ester group thereafter.
