The invention describes a process for the production of a human cartilage implant from chondrocytes cultured in vitro, which come as close as possible to the original with respect to their biochemical composition and biomechanical properties. Up to 20% by vol. of human serum are used as medium addition in the process. The chondrocytes can be kept in monolayer culture until the 12th passage in order firstly to be re- differentiated under a reduced oxygen partial pressure and subsequently to be stimulated to form a three-dimensional cartilage tissue due to aggregation under an oxygen partial pressure of 21%.
L'invention porte sur un procédé permettant de produire un implant de cartilage humain constitué de chondrocytes cultivés in vitro, lequel se rapproche dans la mesure du possible du cartilage d'origine pour ce qui est de la composition biochimique et des propriétés biomécaniques. On a ajouté un volume pouvant aller jusqu'à 20 % de sérum humain dans le milieu de culture au cours de la production. On a pu conserver les chondrocytes en une seule couche jusqu'au 12e passage, d'une part pour subir une deuxième différentiation dans des conditions de pression partielle réduite en oxygène et d'autre part, pour stimuler la formation d'un cartilage tridimensionnel attribuable à l'agrégation des cellules sous une pression partielle en oxygène de 21 %.
