The invention provides a method of rearranging a compound of formula (I), wherein R and R' are selected independently from hydrogen and C1-12alkyl; and R1 and R2 are selected independently from hydrogen, hydroxy, halo, C1-12alkyl- or aryl carbonate, amino, mono- or di-C1-12alkylamino, C1-12alkyl- or arylamido, C1-12alkyl- or arylcarbonyl, C1-12alkyl- or arylcarboxy, C1-12alkyl- or arylcarbamoyl, C1-12alkyl, C2-12alkenyl, C2-12alkynyl, aryl, heteroaryl, C1-12alkoxy, aryloxy, azido, C1-12alkyl- or arylthio, C1-12alkyl- or arylsulfonyl, C1-12alkyl- or arylsilyl, C1-12alkyl- or arylphosphoryl, and phosphato; to form a compound of formula (II), wherein R, R', R1 and R2 are as defined for formula (I); said method comprising treating the compound of formula (I) with a palladium (0) catalyst and a (diphenylphosphino)nC1-6 alkane, wherein n is an integer of 1-6. The invention also provides methods of making penciclovir and famciclovir using this rearrangement reaction.

L'invention concerne un procédé de transformation d'un composé représenté par la formule (I) dans laquelle R et R' sont sélectionnés indépendamment dans hydrogène et alkyle C¿1?-C¿12?; R¿1? et R¿2? sont sélectionnés indépendamment dans hydrogène, hydroxy, halo, carbonate d'aryle ou d'alkyle C¿1?-C¿12?; amino, mono- ou dialkylamino C¿1?-C¿12?, alkyl- ou arylamido C¿1?-C¿12?, alkyl- ou arylcarbonyle C¿1?-C¿12?, alkyl- ou arylcarboxy C¿1?-C¿12?, alkyl- ou arylcarbamoyle C¿1?-C¿12?, alkyle C¿1?-C¿12?, alkényle C¿2?-C¿12?, alkynyle C¿2?-C¿12?, aryle, hétéroaryle, alkoxy C¿1?-C¿12?, aryloxy, azido, alkyl-ou arylthio C¿1?-C¿12?, alkyl-ou arylsulfonyle C¿1?-C¿12?, alkyl- ou arylsilyle C¿1?-C¿12?, alkyl- ou arylphosphoryle C¿1?-C¿12? et phosphato; afin d'obtenir un composé représenté par la formule (II) dans laquelle R, R', R¿1? et R¿2? possèdent la même définition que dans la formule (I); ledit procédé consistant à traiter le composé représenté par la formule (I) au moyen d'un catalyseur de palladium (0) et d'un (diphénylphosphino)¿n?alcane C¿1?-C¿6?, n étant un entier de 1 à 6. Elle concerne également des procédés servant à préparer penciclovir et famciclovir au moyen de cette réaction de transformation.
