The present invention involves a process of purifying and recovering a recombinant protein from a suspension of cells. The process of the present invention involves extracting a recombinant protein from a concentrated suspension of cells using a water-miscible organic solvent, isolating the recombinant protein from the suspension of cells, concentrating the recombinant protein to remove the water-miscible organic solvent, precipitating the recombinant protein using an acid, washing the recombinant protein with the salt or free form of a suitable organic acid and recovering the recombinant protein. The method is particularly applicable to the recovery and purification of recombinant beta-casein.
L'invention se rapporte à un procédé de purification et de récupération d'une protéine recombinante depuis une suspension de cellules. Le procédé de la présente invention consiste à extraire une protéine recombinante depuis une suspension concentrée de cellules au moyen d'un solvant organique miscible avec l'eau, à isoler la protéine recombinante de la suspension de cellules, à concentrer la protéine recombinante pour évacuer le solvant organique miscible avec l'eau, à faire précipiter la protéine au moyen d'un acide, à recombiner la protéine avec le sel ou une forme libre d'un acide organique approprié et à récupérer la protéine recombinante. Ce procédé est tout particulièrement applicable à la récupération et à la purification de la bêta-caséine recombinante.
