Purification of synthesized oligonucleotides is achieved using novel processes and reagents. The novel processes involve adding certain salts to treated synthesized oligonucleotides prior to eluting the oligonucleotides from reverse phase purification cartridges. The novel processes also involve treating synthesized oligonucleotides using cleaving and deprotecting reagents containing methylamine and certain salts. Reagents comprising methylamine and certain salts can be used to rapidly cleave and deprotect oligonucleotides while obtaining high yields of purified oligonucleotides using reverse phase purification technologies.
Purification d'oligonucléotides synthétiques au moyen de processus et de réactifs nouveaux. Les processus nouveaux consistent à ajouter des sels aux oligonucléotides synthétiques traités avant l'éluation des oligonucléotides purifiés par filtration sur cartouche en phase inverse. Ils consistent aussi à traiter les oligonucléotides synthétiques avec des réactifs de clivage et de déprotection contenant de la méthylamine et certains sels. Ces réactifs peuvent être utilisés pour détacher rapidement et déprotéger les oligonucléotides, tout en obtenant des rendements élevés d'oligonucléotides purifiés à l'aide des technologies de purification en phase inverse.
