A prodrug with the use of an enzyme showing different enzymatic activities between a drug target site and a site where its side effect is expressed, which has a substituent cleavable with the enzyme and is activated due by cleaving the substituent. As an example of the drug target site, a respiratory organ may be cited, while the heart may be sited as an example of the side effect expression site. As an example of the drug, a bronchodilator may be cited while a glicosidase (for example, .beta.-glucuronidase) may be cited as an example of the enzyme. As an example of the substituent, a glycosyl group comprising a monosaccharide or an oligosaccharide may be cited. Use of the above enzyme makes it possible to lessen the side effect of a drug having a target site for exerting its effect which is different from a site where its side effect is expressed.
La présente invention porte sur un promédicament qui comprend l'utilisation d'une enzyme présentant des activités enzymatiques différentes entre un site cible du médicament et un site où son effet secondaire est exprimé, ledit promédicament comprenant un substituant clivable par l'enzyme et étant activé par le clivage dudit substituant. On peut citer, à titre d'exemple du site cible du médicament, un organe respiratoire, alors que le coeur peut être un exemple du site d'expression de l'effet secondaire. Comme médicament on peut considérer un bronchodilatateur alors qu'une glycosidase (par exemple, une .beta.-glucuronidase) peut être l'enzyme. Comme exemple pour le substituant, un groupe glycosyle comprenant un monosaccharide ou un oligosaccharide peut être cité. L'utilisation de ladite enzyme permet de réduire l'effet secondaire d'un médicament ayant un site cible d'exercice de son effet qui est différent d'un site où s'exprime son effet secondaire.
