A bioactive compound, or family of compounds, is biosynthesised by a cell using a plurality of enzymic activities. We provide host cells that substantially lack the bioactivity, but possess at least a first one of the enzymic activities (or nucleic acid encoding a corresponding enzyme). The cells are transformed with nucleic acid expressible to provide at least a second enzymic activity which enables the cells to produce a bioactive compound. A family of cells may be transformed with a multiplicity of different nucleic acids, leading to a library of cells including cells producing different bioactive product, and cells not producing bioactive products. Transformed cells may be screened for bioactivity. Active cells may be isolated and cultured, and bioactive compounds may be isolated.

On effectue la biosynthèse d'un composé ou d'une famille de composés bioactifs par une cellule utilisant une pluralité d'activités enzymatiques. L'invention concerne des cellules hôtes manquant pratiquement de cette bioactivité mais possédant au moins une première de ces activités enzymatiques (ou un acide nucléique codant un enzyme correspondant). Ces cellules sont transformées par un acide nucléique dont l'expression permet d'obtenir au moins une deuxième activité enzymatique permettant à ces cellules de produire un composé bioactif. Une famille de cellules peut être transformée par une multiplicité de différents acides nucléiques, ce qui permet d'obtenir une banque de cellules contenant des cellules produisant différents produits bioactifs et des cellules ne produisant aucun produit bioactif. On peut cribler les cellules transformées afin de rechercher leur bioactivité. On peut isoler et cultiver des cellules actives et isoler des composés bioactifs.
