An economical and expedient method is disclosed for the preparation of .alpha.- hydroxy-carboxylic acids or salts thereof in very high enantiomeric purity which comprises oxidizing a corresponding .alpha.-amino-carboxylic acid or salt thereof using an amino acid deaminase followed by reducing the corresponding .alpha.-keto-carboxylic acid or salt produced using a D- or L- lactate dehydrogenase in the combination with an electron donor and an enzyme/substrate system for recycling the electron donor. The resulting .alpha.-hydroxy-carboxylic acids, hydrates, and salts thereof are valuable components and intermediates in the preparation of chiral compounds, especially pharmaceuticals. This invention also relates to the use of .alpha.- amino-carboxylic acids, hydrates, and salts thereof and a coupled enzyme system in the production of .alpha.-hydroxy-carboxylic acids, hydrates, and salts thereof.
L'invention concerne un procédé économique et pratique permettant de préparer des acides .alpha.-hydroxy-carboxyliques ou des sels de ces derniers avec une pureté énantiométique trés élevée. Ce procédé consiste à oxyder un acide .alpha.-amino-carboxylique correspondant ou un sel de ce dernier à l'aide d'une désaminase d'acide aminé, puis à réduire l'acide .alpha.-céto-carboxylique correspondant ou un sel de ce dernier produit à l'aide d'une D- ou L- lactate déshydrogénase en association avec un donneur d'électrons et un système d'enzyme/substrat pour recycler le donneur d'électrons. Les acides .alpha.-hydroxy-carboxyliques, les hydrates, et les sels de ces derniers sont des composants et des produits intermédiaires trés utiles dans la préparation de composés chiraux, en particulier, de produits pharmaceutiques. La présente invention concerne l'utilisation d'acides .alpha.-amino-carboxyliques, d'hydrates, et de sels de ces derniers et d'un système d'enzyme couplé utilisés dans la production d'acides .alpha.-hydroxy-carboxyliques, d'hydrates, et de sels de ces derniers.
