A method to produce a cell expressing an antibody from a genomic sequence of a cell which has a modified immunoglobulin locus is disclosed. The method takes advantage of Cre-mediated site-specific recombination and homologous recombination. The method involves first introducing a lox site into an immunoglobulin locus and then using the lox site as a target for introduction of gene modifications via Cre-mediated in vivo recombination.

Méthode de production d'une cellule exprimant un anticorps à partir d'une séquence génomique d'une cellule comprenant un locus d'immunoglobuline modifiée. Le procédé consiste à exploiter la recombinaison spécifique à un site par Cre ainsi que la recombinaison homologue. Il consiste à d'abord introduire un site lox dans un locus d'immunoglobuline et à utiliser ledit site comme cible pour l'introduction de modifications géniques par recombinaison in vivo induite par Cre.
