Materials and methods for producing fibrinogen in transgenic non-human mammals are disclosed. DNA segments encoding A.alpha., B.beta. and .gamma. chains of fibrinogen are introduced into the germ line of a non-human mammal, and the mammal or its female progeny produces milk containing fibrinogen expressed from the introduced DNA segments. Non-human mammalian embryos and transgenic non-human mammals carrying DNA segments encoding heterologous fibrinogen polypeptide chains are also disclosed.
On décrit des éléments et des procédés qui permettent de produire du fibrinogène chez des mammifères transgéniques autres que l'homme. Des segments d'ADN, codant les chaînes A.alpha., B.beta. et .gamma. du fibrinogène, sont introduits dans la lignée germinale d'un tel mammifère et celui-ci, ou sa descendance femelle, produit du lait contenant la fibrinogène exprimé à partir des segments d'ADN indroduits. On décrit aussi de tels mammifères, ou leurs embryons, dotés de segments d'ADN codant des chaînes polypeptidiques de fibrinogène hétérologue.
