2089752 9204441 PCTABS00011 Human lysozyme (HLZ), a naturally occurring, relatively short, single chain polypeptide, is prepared by growing methylotrophic yeast transformants containing in their genome at least one copy of a first expression cassette comprising a DNA sequence operably encoding HLZ, in operational association with a DNA sequence encoding the S. cerevisiae alpha mating factor pre-pro secretion signal sequence (including the proteolytic processing site: lys-arg), and optionally further including a second expression cassette, comprising a DNA sequence operably encoding HLZ, in operational association with a DNA sequence encoding the native human lysozyme, secretion signal sequence, wherein both the coding sequence(s) and the secretion signal sequence(s) are maintained under the regulation of promoter region(s) of gene(s) of methylotrophic yeast, under conditions allowing expression of said DNA sequences, and secretion of HLZ into the culture medium. Also disclosed are novel DNA fragments and novel recombinant yeast strains which are useful in the practise of the present invention.
