A method is provided for producing motif-specific, context-independent antibodies which are specific to at least one fixed amino acid residue whether modified or unmodified, in the context of variable surrounding amino acid or peptide sequences. The method includes the steps of (1) constructing a peptide library featuring at least one fixed amino acid and variable surrounding amino acids, and (2) immunizing a host with this peptide library. Antibodies may optionally be isolated and purified from the antisera of this immunized host. The disclosed method encompasses both modified motifs, such as phosphothreonine, phosphoserine, MAPK consensus recognition sites, 14-3-3 consensus recognition sites, CDK consensus recognition sites, and acetylated-lysine, as well as unmodified motifs.
L'invention concerne une technique de production d'anticorps spécifiques d'un motif et indépendants du contexte, qui sont spécifiques d'au moins un résidu d'acides aminés fixes, modifié ou non, dans le cadre de séquences de peptides ou d'acides aminés environnants variables. Cette technique comprend les étapes de (1) construction d'une banque de peptides avec au moins un acide aminé fixe et des acides aminés environnants variables, et (2) d'immunisation d'un hôte à cette banque de peptides. Il est possible facultativement d'isoler et de purifier des anticorps à partir des anti-sérums de cet hôte immunisé. Ladite technique comprend des motifs modifiés, tels que la phosphothréonine, la phosphosérine, des sites de reconnaissance de consensus MAPK, 14-3-3, CDK, et la lysine acétylée, ainsi que des motifs non modifiés.
