Plasmids which comprise: (1) an origin of replication; (2) an additional sequence required for plasmid replication or preferably a gene giving a selective advantage; and (3) two expression cassettes each of which are located between (1) and (2) but are separated by (1) and (2) from each other; and which are free from inverted repeat sequences (other than in (3)) are highly persistent though successive generations of microbes containing them.

L'invention concerne des plasmides qui contiennent: (1) une origine de réplication; (2) un séquence supplémentaire requise pour la réplication des plasmides ou de préférence un gène conférant un avantage sélectif, (3) deux cassettes d'expression situées chacune entre (1) et (2) mais séparées l'une de l'autre par (1) et (2). Lesdits plasmides sont de plus exempts de séquence répétées inverses (autres que celle dans (3)) et sont hautement persistants malgré les générations de microbes successives les contenant.
